What does an O mean at the beginning of an MLS number?

Megan,

The "O" represents the designated area "multiple listing service" the property is listed on. When looking up a specific property it is necessary to enter both the letter and the numbers following it to locate the property.

Nadine is right. THe Letter in from of the MLS# refers to the county the listing originated from. In this case "O"
refers to Orlando.

An "O" means its a condo. :)

It's listed by the Orlando Board of Realtors.
