One can pick up a Trans-Bridge bus at either the Trans-Bridge terminal off of Schoenersville Road and Route 22 or at the "Park and Ride" located at Route 33 and William Penn Highway. One needs to purchase the bus ticket (monthly is easier) at the Trans-Bridge terminal itself. In theory, the commute to either the Port Authority or the Financial Center is an hour and a half. In reality, it is usually closer to two hours.
If you were to drive yourself, depending on where in Manhattan you need to commute to, it would take between two to two and a half hours.
There is no train service from the Lehigh Valley to Manhattan. Your best option would be to drive to a New Jersey Transit station that has fairly frequent service, such as Summit, and take the train from there. That would take you to Hoboken, from where you could get either a Path train or a ferry over to Manhattan.
